ThedefinedantibodyreactivityisrestrictedtoFXI,FXIaandcomplexesofFXIwithHMWKorotherproteins.Inimmunoelectrophoresis,bi-dimensionalelectrophoresisandrADIalimmunodiffusion(Ouchterlony)againstnormalplasma,asingleprecipitinlineisobtainedwhichshowsareactionofidentitywithprecipitatedpurifiedFactorXI.NoreactionisobtainedwithFXI-depletedplasma.ThisantiserumisprimarilyintendedforthemeasurementofFactorXIinhumanplasmausingtheEIDmethod(Laurell).FXIisoneofthefourproteinsknowntobeinvolvedinnormalcontactactivationreactions,activatingFXIItoFXIIa,whichisfurthercapableoftriggeringthekinin-formingpathwayandthefibrinolyticsystem.FXIispresentinnormalhumanplasmabothinfreeformandasareversIBLecomplexwithHMWK.FXIcompeteswithprekallikrein(PKK)toformsuchcomplexes.SinceFXIisagammaglobulinwithcathodicmobilityinelectrophoresis,upto140_g/mlpurifiedHMWKmustbeaddedtotheplasmatobindallfreeFXIintonegativelychargedcomplexeswhichwillmigratetotheanode.ThetotalFXIcontentcanthenbemeasured.TheaverageamountofFXIinanindividualplasmawasfoundtobe4.5_g/mlwitharangeof3to6_g/ml.ThesensitivityoftheEIIDmethodisabout0.2_g/ml.AcongenitaldeficiencyofFXIinplasmacangiverisetoaseriousbleedingdisorder.InmostcasestheplasmalevelofFXIantigenisalsodepressed;exceptionallytheFXIantigenisnormal(adefectivemolecule)AnacquiredreductioninbothclottingactivityandFXIantigenlevelhasbeendescribedindisseminatedintravascularcoagulopathy,inhepaticcirrhosisandinsomepatientswithlupuserythomatosus.
